Alzheimer’s disease (AD) is defined by the misfolding and accumulation of Aβ peptides and of tau proteins in brain parenchyma. The most frequent clinical presentation of sporadic AD is a progressive cognitive decline with a mean disease duration of 8 years. However, the phenotypic heterogeneity is well known, including a rapidly progressive subtype of AD (rpAD).
Although the molecular basis of the phenotypic diversity in AD is still unknown, it is reminiscent of a strain-like phenomenon as described in prion diseases. In recent years, the seeding activity of Aβ and tau have been repeatedly demonstrated using transmission paradigms following brain inoculation. It has been seen that the propagation of distinct tau strains in cells and mice define different tauopathies.
We are interested in studying a group of patients which are characterized by a rapidly progressive development of AD, that is frequently misdiagnosed as Creutzfeldt-Jakob disease. Our team has already contributed to describe, in a multicentric study, specific traits associated with rpAD not observed in standard AD cases with slower progression. The short duration of the clinical phase and the fast cognitive decline of rpAD suggest a potential aggressive strain that we plan to isolate by transmission experiments in mice.
We are seeking a highly motivated postdoctoral fellow with a Ph.D. in neuroscience, neurobiology, or a related field. Preference will be given to candidates with experience in mouse stereotaxic injections, behavioral assessment and histological analysis, however highly motivated candidates with alternate experience are also encouraged to apply. The successful candidate will work with human brain tissue in specialized infrastructures and will benefit from the support of a multidisciplinary team (Susana Boluda, MD, PhD; Benoît Delatour, PhD) with expertise in neuropathology, animal neurobiology, biochemistry and experimental transmission in the field of AD and prion diseases.
