The group of Prof. Christian P. Müller at the Psychiatric and Psychotherapeutic Clinic of the University Hospital Erlangen-Nuremberg (FAU) offers a PhD position with a thematic focus on the study of the effects and loss of efficacy of antipsychotic drugs in the treatment of schizophrenia. The project investigates how the observed accumulation of different antipsychotics in cellular vesicles during treatment (Tischbirek et al., Neuron, 2014) leads to the often observed loss of efficacy of the therapy and how to prevent it. By means of various non-accumulating substance analogues, a rat model of schizophrenia will be used to investigate how the accumulation of antipsychotics first leads to therapeutic effects over several symptom dimensions (psychosis, emotionality and cognition) (Uzuneser et al., Eur. Neuropsychopharm. 2018). Furthermore, we will measure how the accumulation develops after chronic administration and which of the subsequent cellular adaptation processes leads to the loss of efficacy in each symptom dimension. Thereafter, new therapeutic interventions are to be tested, which can specifically prevent the loss of efficacy (Amato et al., Molecular Psychiatry, 2018). Our laboratory has in-depth expertise in neurochemical, behavioral and molecular biology techniques related to the study of schizophrenia in animal models, as well as a long history in the study of the effects of neurochemical interventions.
