A Post-Doctoral position is available at the University of Versailles Saint Quentin en Yvelines in the team “Biotherapies of neuromuscular diseases” at U1179 INSERM-UVSQ.
The project aims at evaluating new therapeutic approaches to restore the expression of various dystrophin isoforms in the brain of DMD mouse models and assessing the extent of the behavioural impact following this postnatal dystrophin restoration.


Context and Job description:
Duchenne muscular dystrophy (DMD) is the most common muscular dystrophy in childhood and is also associated with cognitive deficits that can lead to intellectual disability and neuropsychiatric disorders (autism, ADHD, TOCs). The nature and severity of the cognitive dysfunctions and brain alterations vary with the position of the mutations in the DMD gene, which may differently affect the expression of several brain forms of dystrophin.
Our multidisciplinary and collaborative research program aims to clarify the genotype-phenotype relationships in this disease and develop antisense therapeutic approaches to correct the DMD gene and restore the expression of functional brain dystrophins.
The Postdoctoral candidate will develop different antisense tools (antisense oligonucleotides and AAV vectors) targeting different DMD exons to restore the expression of various brain dystrophin isoforms and assess the impact that postnatal restoration of dystrophin expression has on neuropsychological /behavioural features in DMD mouse models.

Profile:
The applicants should have a PhD in neuroscience, genetics or a closely related field. Good knowledge of neurobiology and biotherapies will be required, as well as a strong experience in molecular biology (RNA biology, qPCR, WB, IF, In situ hybridization) and in vivo work in mice. Previous experience with antisense therapeutics and/or AAV viral vectors would be appreciated.
